Human LILRA3, also known as Leukocyte Ig-like receptor 4 (LIR-4), CD85 antigen-like family member E, Immunogobulin-like transcript 6 (ILT-6) and monocyte inhibitory receptor HM43/HM31, is a 70 kD Ig superfamily member that belongs to the leukocyte receptor complex/cluster. Mature LILRA3 is 416 amino acids (aa) in length and contains four C2-type Ig-like domains. Unlike other LILR family members, LILRA3 is actively secreted. Alternative splicing generates additional isoforms with one containing an extra 18 aa in the signal peptide and/or a 64 amino acid deletion in the C-terminal half of the second Ig-like domain. Human LILRA3 shares ~86% aa identity with Chimpanzee LILRA3. Human LILRA3 acts as a soluble receptor for class I MHC antigens. It binds both classical and non‑classical HLA class I molecules but with reduced affinities compared to LILRB1 or LILRB2.
