一、家族规模与命名
TNF受体超家族(TNF Receptor Superfamily, TNFRSF)目前包含29个成员,编号从TNFRSF1A至TNFRSF29。其配体属于TNF超家族(TNFSF),包含19个成员。这些分子形成复杂的调控网络,贯穿免疫系统的发育、活化、记忆形成和稳态维持。
(1)结构特征
所有TNFR家族成员共享以下结构特点:
胞外域:1-6个富含半胱氨酸的结构域(CRD),形成特征性的"CRD棒"构象
跨膜域:单次跨膜α螺旋
胞内域:长度从4个氨基酸(TACI)到超过400个氨基酸(NGFR)不等。
(2)根据胞内域结构分为两类:
死亡受体:含死亡结构域(DD),包括TNFR1、Fas、DR3、DR4、DR5、DR6,主要诱导细胞凋亡或坏死性凋亡
非死亡受体:无DD,通过TRAF蛋白传递信号,包括TNFR2、CD40、CD27、4-1BB、OX40、GITR等,主要促进细胞存活和活化

(4)配体-受体相互作用特点
TNFR家族配体-受体结合具有以下特征:
a. 三聚体配体:多数配体以同源三聚体形式存在,交联三个受体分子。
b. 受体预聚集:部分受体(如4-1BB、TNFR1)在细胞表面预先形成二聚体或寡聚体,配体结合后进一步稳定聚集。
c. 反向信号:部分配体(如CD40L、4-1BBL、FasL)可向表达细胞传递反向信号。
d. 可溶型调控:金属蛋白酶(ADAM10/17、TACE)切割产生可溶型配体,通常活性降低或表现为拮抗剂。
e. 诱骗受体:部分受体(如DcR1、DcR2、OPG)缺乏功能性胞内域或作为可溶型存在,竞争性结合配体但阻断信号传导。
