T细胞表面糖蛋白CD28（又称TP44）是一种I型跨膜蛋白，含有一个免疫球蛋白样V型结构域。它提供T细胞活化所需的共刺激信号，是CD80（B7.1）和CD86（B7.2）的受体。抗原呈递细胞（APC）活化后上调CD80表达，而CD86呈组成性表达。CD28是初始T细胞上唯一组成性表达的B7受体。
