Catechol-O-methyltransferase catalyzes the transfer of a methyl group from S-adenosylmethionine to catecholamines, including the neurotransmitters dopamine, epinephrine, and norepinephrine. This O-methylation results in one of the major degradative pathways of the catecholamine transmitters. In addition to its role in the metabolism of endogenous substances, COMT is important in the metabolism of catechol drugs used in the treatment of hypertension, asthma, and Parkinson disease. COMT is found in two forms in tissues, a soluble form (S-COMT) and a membrane-bound form (MB-COMT). The differences between S-COMT and MB-COMT reside within the N-termini. Several transcript variants are formed through the use of alternative translation initiation sites and promoters.

Flow cytometric analysis of Catechol-O-methyltransferase expression in HT-1080 cells using Catechol-O-methyltransferase antibody . Green, isotype control; red, Catechol-O-methyltransferase.

Immunocytochemical staining of HT-1080 cells with Catechol-O-methyltransferase antibody (Cat #61443, 1:1,000). Nuclei were stained blue with DAPI; Catechol-O-methyltransferase was stained magenta with Alexa Fluor® 647. Images were taken using Leica stellaris 5. Western blotting analysis using Catechol-O-methyltransferase antibody . Total cell lysates (30 μg) from various cell lines were loaded and separated by SDS-PAGE. The blot was incubated with Catechol-O-methyltransferase antibody and HRP-conjugated goat anti-rabbit secondary antibody respectively. Image was developed using FeQ™ ECL Substrate Kit .
