Abstract
Metabarcoding continues to be a valuable tool for exploring soil nematode diversity and its ecological implications. However, ecological studies often entail large-scale sampling, necessitating significant time for transportation and analysis. Therefore, ensuring the preservation of DNA integrity is crucial for successful metabarcoding research. While various chemicals have been developed for soil and tissue preservation, with tests conducted on various organisms, limited work has focused on nematodes. In our study, we investigated the influence of sample preservation methods on nematode community recovery. Although the diversity recovered from ethanol-preserved soil exhibited minimal changes over time, a lower proportion of nematode taxa was detected, indicating that soil preservation is not ideal. For extracted nematodes, preservation in ethanol proved more effective than DESS. We recommend using freshly extracted nematodes for ecological studies, as all preservation methods can alter community composition. When preservation is necessary, storing extracted nematodes in ethanol for a maximum of 7 days has a lesser impact on community composition.
