DEAD box proteins, characterized by the conserved motif Asp-Glu-Ala-Asp (DEAD), are putative RNA helicases which are implicated in a number of cellular processes involving RNA binding and alteration of RNA secondary structure. This gene encodes a protein containing RNA helicase-DEAD box protein motifs and a caspase recruitment domain (CARD). It is involved in viral double-stranded (ds) RNA recognition and the regulation of the antiviral innate immune response. Mutations in this gene are associated with Singleton-Merten syndrome 2.

Western blotting analysis using RIGI antibody . Total cell lysates (30 μg) from various cell lines were loaded and separated by SDS-PAGE. The blot was incubated with RIGI antibody and HRP-conjugated goat anti rabbit secondary antibody respectively. Image was developed using FeQTM ECL Substrate Kit .

Western blotting analysis using RIGI antibody . RIGI expression in wild-type (WT) and RIGI shRNA knockdown (KD) HSHC cells with 20 μg of total cell lysates. Hsp90 α serves as a loading control. The blot was incubated with RIGI antibody and HRP-conjugated goat anti-rabbit secondary antibody respectively. Image was developed using NaQ™ ECL Substrate Kit .

Validation of RIGI knockdown using flow cytometry. Wild-type(WT, Blue) and knockdown(KD, Green) HAP1 cells were stained with RIGI antibody and analyzed using BD flow cytometer.
